How to control the immersion depth and angle of manual pipette tips?

Controlling the immersion depth and angle of manual pipette tips is a critical step in ensuring accurate aspiration of liquids. Here are some suggestions:
Control of immersion depth:
The immersion depth should ensure that the required pipetting volume is achieved while minimizing contact of the outer wall of the tip with the liquid.
Choose the appropriate immersion depth based on the pipette specifications. For example, for tips below 10 μl, the immersion depth is usually 1~2mm; for tips above 2000μl, the immersion depth is 6~10mm.
When pipetting large volumes or liquids with high viscosity, special attention should be paid to the control of immersion depth to ensure that the liquid is fully sucked into the tip.
Control of immersion angle:The angle at which the tip is immersed in the liquid should be controlled within 20° and kept as vertical as possible. This helps reduce problems with too much or too little liquid intake due to excessive angles.
In actual operation, the immersion angle can be adjusted by observing the contact between the tip and the liquid surface to ensure that the tip can enter the liquid smoothly and accurately.
In addition, you need to pay attention to the following points:
Before aspirating liquids, you can gently rinse the tips to help improve pipetting accuracy. However, for high or low temperature samples, rinsing may reduce accuracy, so judgment needs to be made based on actual conditions.
When aspirating liquids, maintain a steady aspiration speed and avoid being too fast or too slow. Too fast aspiration speed may cause the sample to enter the handle, causing damage to the piston and sealing ring and cross-contamination of the sample.
After absorbing the liquid, keep the tip in the liquid surface for a period of time (such as 1 to 3 seconds), and then gently move the tip away from the liquid surface. This helps ensure that liquid is fully drawn into the tip and reduces dripping or splashing caused by moving the tip.
